Research Overview

CJC-1295 / Ipamorelin Blend (no DAC) (Research Overview)

This product is a blend of two separate peptides: CJC-1295 without the drug affinity complex (DAC), a modified analogue of growth hormone-releasing hormone fragment 1-29, and ipamorelin, a synthetic pentapeptide (Aib-His-D-2-Nal-D-Phe-Lys-NH2) classed as a growth hormone secretagogue. It is not the same material as CJC-1295 with DAC, which is a separate product.

Mechanism of Action

The two components act at different G protein-coupled receptors. Growth hormone-releasing hormone (GHRH) analogues engage the GHRH receptor, which signals through cAMP, and the neuroendocrine control of growth hormone involves interplay between GHRH and somatostatin neurons. Reviews describe the receptor, its splice variants and its signalling pathways.

Ipamorelin was described in 1998 as a pentapeptide derived from a series of compounds related to growth hormone-releasing peptide (GHRP)-1, and pharmacological profiling with receptor antagonists indicated that it acts through a GHRP-like receptor. That receptor, the growth hormone secretagogue receptor, was cloned in 1996, and an endogenous acylated peptide ligand, ghrelin, was identified in 1999.

Research Areas

Receptor pharmacology: GHRH receptor agonists and growth hormone secretagogue receptor agonists are characterized in separate cell-based and animal systems, where potency and receptor selectivity are compared.

Neuroendocrine background: reviews of growth hormone regulation describe the hypothalamic and pituitary circuitry involved in the two receptor pathways.

Component identity: because the two peptides have distinct structures and receptors, studies of either one individually (see the CJC-1295 and Ipamorelin research overviews) are the primary literature for each component.

Findings from published studies do not establish safety or efficacy for any indication and should not be interpreted as medical claims.
