Research Overview

DSIP (Delta Sleep-Inducing Peptide) (Research Overview)

Delta sleep-inducing peptide (DSIP) is a nonapeptide with the sequence Trp-Ala-Gly-Gly-Asp-Ala-Ser-Gly-Glu and a molecular weight of about 849 Da. It was isolated from rabbit cerebral venous blood and characterized in a 1977 report that gave the peptide its name.

Mechanism of Action

In the original characterization, the isolated peptide and a set of synthetic analogues and fragments were synthesized and compared by electroencephalogram analysis after intraventricular infusion in rabbits, and only the synthetic nonapeptide showed the specific change in EEG activity that led to its name. The structure of DSIP is unlike that of any other known peptide family.

Later reviews note that the gene and protein precursor of DSIP and a dedicated receptor have not been identified, that its natural occurrence and biological activity remain unclear, and that some investigators have proposed the existence of one or more DSIP-like peptides to account for the immunoreactivity and activity reported. The mechanism of action of DSIP is therefore described in the literature as unresolved.

Research Areas

Neuropeptide history and characterization: DSIP is one of a small number of peptides purified to homogeneity and chemically characterized in the search for endogenous factors in blood and brain, and the 1977 report remains the primary reference for its structure.

Literature on distribution and function: reviews summarize immunochemical and radioimmunochemical work on DSIP-like material in brain and peripheral tissue and the range of physiological parameters examined in animal studies.

Open questions: a recent mini-review discusses why the identity and natural activity of DSIP remain uncertain and what would be needed to resolve them.

Findings from published studies do not establish safety or efficacy for any indication and should not be interpreted as medical claims.
