Research Overview

TB-500 (Research Overview)

TB-500 is a synthetic analog of the active region of Thymosin Beta-4 (Tβ4), a naturally occurring 43-amino acid peptide. Research has focused on its role in actin regulation, angiogenesis, and systemic tissue repair signaling.

Mechanism of Action

TB-500 corresponds to the actin-binding domain of Thymosin Beta-4 (amino acids 17–23, sequence LKKTETQ). Its primary studied mechanism involves sequestration of G-actin monomers, which regulates actin polymerization dynamics critical for cell migration and tissue remodeling. This actin-regulatory function is thought to underlie many of its observed effects on wound healing and repair processes.

Research has also examined TB-500's effects on upregulation of cell surface receptors, promotion of angiogenesis through VEGF pathway interactions, and modulation of inflammatory cytokine profiles. Its systemic distribution — unlike many peptides that act locally — has been a notable feature in preclinical studies.

Research Areas

Musculoskeletal repair research has been a primary focus, with studies examining TB-500's effects on muscle fiber regeneration, tendon healing, and cardiac muscle repair following ischemic injury. The cardiac research context has been particularly active, with multiple groups investigating Thymosin Beta-4 and its analogs for post-infarction myocardial repair.

Neurological research represents a growing area, with preclinical studies examining TB-500's effects on oligodendrocyte differentiation, remyelination, and neuroprotection following CNS injury. Anti-inflammatory properties have been studied in models of chronic inflammation and autoimmune conditions.

TB-500 is frequently studied in combination with BPC-157, as the two peptides are thought to act on complementary repair pathways — BPC-157 addressing local tissue repair and TB-500 providing systemic healing support. This combination is available as a blend in our catalog.
