Research Overview

5-Amino-1MQ (Research Overview)

5-Amino-1MQ is the common name for 5-amino-1-methylquinolinium, a small organic molecule rather than a peptide. It belongs to the family of N-methylated quinolinium compounds that have been studied as inhibitors of the enzyme nicotinamide N-methyltransferase (NNMT).

Mechanism of Action

NNMT is a cytosolic enzyme that transfers a methyl group from the cofactor S-adenosyl-L-methionine (SAM) to nicotinamide, a form of vitamin B3, and to other pyridine-containing compounds. It was first characterized as an enzyme of nicotinamide clearance, and later work has examined its role in regulating multiple metabolic pathways through the consumption of methyl donors and the generation of methylated products.

Structure-activity studies screened N-methylated quinolinium, isoquinolinium, pyridinium and benzimidazolium or benzothiazolium analogues and identified the quinolinium scaffold as an NNMT inhibitor series with low-micromolar potency in vitro, supported by docking to the nicotinamide-binding site. Separate biochemical and crystallographic work has produced a structure of human NNMT in complex with a small-molecule inhibitor.

Research Areas

Enzymology and inhibitor design: NNMT inhibitors, including quinolinium analogues, are used as chemical tools to test hypotheses about the enzyme's role in cells and in biochemical assays.

Structural biology: crystal structures of NNMT with bound inhibitors inform structure-activity relationships and docking models.

Background literature: reviews discuss NNMT at the crossroads of cellular metabolism and epigenetic regulation through its use of SAM, and summarize open questions about its function.

Findings from published studies do not establish safety or efficacy for any indication and should not be interpreted as medical claims.
