Research Overview

ARA-290 (Cibinetide) (Research Overview)

ARA-290, also called cibinetide, is an 11-residue linear peptide designed from the aqueous face of helix B of erythropoietin (EPO). It was developed to separate tissue-protective signaling studied for EPO from EPO's effects on red blood cell production.

Mechanism of Action

Studies describe ARA-290 as interacting with a receptor complex formed by the EPO receptor and the β-common receptor (CD131), sometimes called the innate repair receptor. Unlike EPO, it does not activate the classical EPO receptor homodimer used for erythropoiesis in published assays.

The peptide begins with pyroglutamate, which blocks the N-terminus, and has a listed molecular weight of about 1257 Da.

Research Areas

Preclinical work has used ARA-290 in rodent models of nerve injury, inflammation and metabolic stress, and it has been evaluated in early clinical studies as an investigational compound. It is not an approved drug.

For laboratory characterization, the pyroglutamyl N-terminus and the absence of cysteines are relevant: identity is confirmed by mass spectrometry, and N-terminal sequencing by Edman degradation is blocked.
