Sermorelin (Research Overview)
Sermorelin is a synthetic peptide matching the first 29 residues of human growth hormone-releasing hormone (GHRH), with an amidated C-terminus. The 1–29 fragment is the shortest sequence reported to retain the full receptor activity of the 44-residue native hormone.
Mechanism of Action
Sermorelin binds the GHRH receptor (GHRH-R), a class B G protein-coupled receptor on pituitary somatotroph cells. Receptor activation raises intracellular cyclic AMP, the pathway through which GHRH signaling is studied.
Unmodified GHRH(1–29) is rapidly cleaved by dipeptidyl peptidase-4 and other proteases, giving it a short half-life in biological matrices. Later analogs, such as CJC-1295 and Tesamorelin, were designed with substitutions or added groups to extend that half-life, which makes Sermorelin a common reference compound in comparisons.
Research Areas
Research has used Sermorelin as a reference GHRH-receptor agonist in pituitary cell and animal models, and in studies of GHRH-receptor structure-activity relationships.
Sermorelin was previously marketed in the United States as a drug product that was later withdrawn by its manufacturer. Material sold by Sealed Peptides is for laboratory research use only.
